BeOne Medicines and Revolution Medicines Announce Broad Oncology Collaboration to Advance RAS-Targeted Therapies
BeOne Medicines Ltd. and Revolution Medicines, Inc. have announced a multi-part strategic collaboration aimed at expanding treatment options for patients with RAS-addicted cancers. The agreement combines clinical development efforts around targeted drug combinations with a separate regional licensing arrangement that will give BeOne exclusive rights to develop and commercialize four clinical-stage RAS(ON) inhibitors from Revolution Medicines in selected Asian markets.
The collaboration brings together two oncology-focused companies with complementary strengths. BeOne Medicines contributes an established global research, clinical development and commercial infrastructure, while Revolution Medicines brings a portfolio of targeted RAS(ON) inhibitors designed to address cancers driven by oncogenic RAS signaling. Together, the companies intend to investigate potentially complementary treatment combinations while also expanding access to Revolution Medicines’ RAS-targeted pipeline across important Asian markets.
RAS mutations are among the most common genetic alterations associated with cancer. Because RAS proteins play a central role in transmitting signals that regulate cell growth and survival, abnormal RAS signaling can contribute directly to tumor development and progression. Certain cancers become particularly dependent on these pathways, a characteristic commonly described as “RAS addiction.” Developing medicines that can selectively interfere with active RAS signaling has therefore become an important area of oncology research.
Exploring New Combination Strategies
A central component of the collaboration is a clinical research program designed to evaluate combinations involving selected BeOne Medicines oncology assets and Revolution Medicines’ four clinical-stage RAS(ON) inhibitors.
The Revolution Medicines portfolio included in the collaboration comprises daraxonrasib, a RAS(ON) multi-selective inhibitor; zoldonrasib, a RAS(ON) G12D-selective inhibitor; elironrasib, a RAS(ON) G12C-selective inhibitor; and RMC-5127, a RAS(ON) G12V-selective inhibitor.
These investigational medicines are designed to target different forms of active RAS proteins. The ability to address specific RAS variants could potentially allow researchers to develop more precisely targeted treatment strategies for patients whose tumors are driven by particular RAS alterations.
Under the clinical collaboration, planned combination studies will include BeOne’s MTA-cooperative PRMT5 inhibitor BGB-58067 and its EGFR x MET x MET trispecific antibody BG-T187. These candidates will be evaluated in combination with either daraxonrasib or zoldonrasib.
The companies believe that combining medicines with complementary biological mechanisms could potentially enhance anti-tumor activity. However, the effectiveness and safety of these combinations will need to be established through clinical research.
The collaboration is therefore designed to generate clinical evidence that may help determine whether pairing RAS(ON) inhibition with other targeted approaches can provide additional benefits for patients with difficult-to-treat cancers.
Regional Licensing Agreement Expands Access in Asia
In addition to the clinical collaboration, Revolution Medicines has granted BeOne Medicines exclusive rights in selected Asian markets to develop and commercialize, or solely commercialize depending on the market, the four clinical-stage RAS(ON) inhibitors included in the agreement.
The regional rights provide BeOne with an opportunity to leverage its established development capabilities and commercial presence in Asia to potentially expand access to Revolution Medicines’ RAS-targeted medicines.
Revolution Medicines will remain responsible for development and commercialization of its assets outside the licensed territory. Importantly, Japan and South Korea are excluded from the partnered region, allowing Revolution Medicines to retain its rights in those markets.
The arrangement also creates a financial opportunity for Revolution Medicines. Under the agreement, the company will be eligible to receive development and sales milestone payments as well as tiered royalties based on net sales of the partnered products within the licensed territories.
This structure gives both companies incentives to successfully advance and commercialize the therapies. BeOne gains access to a promising portfolio of targeted oncology assets, while Revolution Medicines gains an experienced regional partner capable of supporting development and commercialization across selected Asian markets.
BeOne to Support Global Phase 3 Development
Another important element of the agreement is BeOne’s commitment to fund and conduct a global registrational Phase 3 study for one of Revolution Medicines’ RAS(ON) inhibitors.
The study will be supported by BeOne’s fully integrated development infrastructure, which the company describes as its differentiated “development superhighway.” This model is intended to allow BeOne to move medicines efficiently through clinical development by leveraging its internal research, regulatory, clinical and commercial capabilities.
While BeOne will take responsibility for the selected global Phase 3 program, Revolution Medicines will continue advancing a broad portfolio of global registrational studies involving its RAS(ON) inhibitors.
The complementary approach allows both companies to pursue multiple development opportunities simultaneously while expanding the overall reach of the RAS-targeted portfolio.
For BeOne, the agreement represents another opportunity to build upon its growing presence in oncology and utilize its global development capabilities. For Revolution Medicines, the partnership provides access to BeOne’s regional infrastructure and creates additional pathways for its RAS(ON) inhibitor portfolio to reach patients.
Addressing Unmet Needs in RAS-Driven Cancers

Although researchers have made progress in developing therapies against certain RAS mutations, many patients continue to have limited treatment options, particularly when their disease progresses or develops resistance to existing therapies.

The ability to investigate these medicines in combination with other targeted therapies could potentially broaden their therapeutic applications and identify new strategies for treating RAS-addicted tumors.
